Dynamics of complex feedback architectures in gene-metabolic pathways

Abstract: The interactions between genetic and metabolic networks display intricate feedback architectures that combine positive and negative regulatory loops. These are fundamental for cell survival, as they are the main mechanism by which cells adapt their metabolism to large environmental fluctuations. Here we present a novel analysis technique that blends ideas from timescale separation and piecewise affine dynamical systems, applied for a wide class of unbranched metabolic pathways under nonlinear genetic regulation. We illustrate our results on a system that closely resembles the "metabolator", a synthetic genetic-metabolic circuit engineered to produce autonomous oscillations in vivo, and prove the existence of two co-existing oscillatory behaviors: damped oscillations towards a fixed point or sustained oscillations along a periodic orbit.
